Study on the Optimal Storage Conditions for Granulocyte Transfusion using MTT Assay

Abstract
Granulocyte transfusions have proven to be efficacious in prevention or treatment for certain types of infections in neutropenic patients. It has been known that granulocytes stored in vitro lose their capacity to circulate when transfused after several hours of storage, therefore ineffective granulocytes transfusion can be resulted using stored granulocytes. The author performed an in vitro study to evaluate the effects of granulocytes storage time and conditions by the measurement of viability using MTT assay and compared that using trypan blue dye exclusion test. . The results are summarized as follows: 1. Among the separated granulocytes stored in ACD plasma, heparin plasma, complete RPMI- 1640 and HBSS, granulocytes stored in ACD plasma showed the highest viability until 72 hour storage(p(t)
